Разработчики не стали придумывать что то новое, ведь сама суть игры, которая основана на мультфильме, в том, чтобы позволить фанатам окунуться в эту атмосферу, которую они с нетерпением ждали каждый раз у экранов телевизора. В основе сюжета 4 главных персонажа: Рафаэль, Донателло, Микеланджело, Леонардо. Разумеется, что как и в мульфильме черепашки стоят на защите города Нью Йорка. Но постоянно им на пути появляются те или иные злодеи. Стоит отметить, что после того, как игрок запустит игру и просмотрит ролик, повествующий о том, для чего и зачем была создана игра, ему будет предоставлен выбор режимов. Разработчики создали два режима: в одном сюжет идет плавно, на протяжении всей игры уровни плавно сменяют друг друга, при этом сложность игры будет увеличиваться. В другом же режиме можно будет просто драться с любыми персонажами. Сам игрок может выбрать черепашку, которая будет драться, а также выбрать злодея, которого нужно будет одолеть. Разумеется, что в обычном режиме, который предусматривает наличие истории, гораздо интереснее. Там огромное количество миссий, с различными заданиями. Также можно менять иногда черепашек. Не стоит забывать о том, что все черепашки различаются друг от друга. У них разные способности, а также разные сильные стороны. Иногда функция, которая позволяет сменить персонажа, довольно эффективная.
